肾苏Ⅱ对系膜增生性肾炎模型大鼠肾病变及白细胞介素-6的影响  

Influence of Shensu-Ⅱ on the Renal Pathological Changes and Interleukin-6 Expression in Mesangium Proliferative Glomerulonephritis Rat Models

摘　　要：目的探讨中药复方肾苏Ⅱ对系膜增生性肾炎(MsPGN)模型大鼠肾组织的作用及机制。方法 40只雄性SD大鼠随机分为正常组、模型组、西药组和中药组,每组10只。其中,除正常组外,其余大鼠采用胸腺细胞抗体注射法诱发MsPGN模型。中药组于造模的同时灌服治疗药物肾苏Ⅱ(3.5g/100g),西药组灌服贝那普利(0.33mg/100g),共干预6周。于治疗开始及治疗第3、6周末,取血观察各组大鼠24h尿蛋白(TP/24h)、尿素氮(BUN)及肌酐(SCr)变化;治疗第6周末,切取肾脏,光镜观察系膜细胞、基质增殖,检测白细胞介素6(IL-6)表达。结果中药组可显著减少MsPGN大鼠尿蛋白排泄、降低BUN及SCr含量,与西药组比较差异有统计学意义(P<0.05);明显抑制系膜细胞及基质增殖与西药组比较差异无统计学意义(P>0.05);显著下调肾组织IL-6表达,与西药组比较差异有统计学意义(P<0.05)。结论肾苏Ⅱ可改善MsPGN模型大鼠肾功能,减轻病理损害,阻抑系膜细胞与基质增殖及肾组织IL-6高表达,延缓病变进展。Objective To explore the effect and mechanism of Shensu-Ⅱ（Chinese medicine for nephrosis）on the renal tissues of rat models of mesangium proliferative glomerulonephritis（MsPGN）.Methods Forty male SD rats were randomized into four groups,i.e.normal group,model group,western medicine group,and Chinese medicine group,with 10 in each.Except the normal group,the rats were made MsPGN models by using the thymus cell antibody injection.The Chinese medicine group was treated with Shensu-Ⅱ 3.5g/100g through intragastric filling,and the western medicine group treated with Benazepril 0.33mg/100g for control.The intervention was of six weeks in total.From the starting of treatment and at the end of the third and sixth week of treatment,the blood samples of all groups were collected for observing the changes in urine protein in 24h（TP/24h）,blood urea nitrogen（BUN）,and serum creatinine（SCr）;at the end of the sixth week of treatment,the kidneys were cut off for observing the proliferation of mesangium cells and matrix with light microscope,and the expression of interleukin-6（IL-6）.Results In the Chinese medicine group,the excretion of urinary protein and content of BUN and SCr of the MsPGN rat models were significantly reduced,and the difference was significant in comparing with those in the western medicine group（P0.05）;the proliferation of mesangium cells and matrix were significantly inhibited,and the result of Masson dyeing image analysis showed that there was no significant difference from that of the western medicine group（P0.05）.IL-6 expression of the renal tissues was significantly down regulated,and the difference was significant in comparing with that in the western medicine group（P0.05）.Conclusion Shensu-Ⅱ is effective to improve the renal function of MsPGN rat models,reduce its pathological damage,and inhibit the proliferation of membrane cells and matrix and the exessive expression of renal IL-6 as well,thus delaying the progression of disease.
