内蒙古2种白刺间自然杂交类型的分子验证

摘　　要：本实验对齿叶白刺(Nitraria roborowiskiiKom.)和唐古特白刺(N.tangutorum Bobr.)以及疑似杂交个体共48个个体的叶绿体trnL-F序列和核糖体ITS序列进行分析.trnL-F结果显示,齿叶白刺和唐古特白刺在15个位点上存在差异,杂交个体在其中11个位点上与齿叶白刺相同,4个位点与唐古特白刺相同,推测齿叶白刺为杂交个体的母本,唐古特白刺为父本.ITS结果显示,齿叶白刺和唐古特白刺在23个位点上存在差异,杂交个体在其中10个位点上与齿叶白刺相同,7个位点与唐古特白刺相同,在其余6个位点上所有杂交个体的ITS序列均出现了位点的杂合.结果表明,中间形态个体是齿叶白刺和唐古特白刺的自然杂交后代;对2个亲本种以及杂交群内个体间trnL-F和ITS序列的变异状况分析认为,可能是杂交群经历了较长时间的繁衍,造成了杂交群体内序列变异的多样化.The sequence comparison of trnL-F intergenic spacer region of the chloroplast DNA and the internal transcribed spacer（ITS） regions of nuclear ribosomal from Nitraria roborowskii,N.tangutorum and the suspected hybrids was performed.The result of trnL-F sequence analysis revealed total 15 variable loci were detected from N.roborowskii and N.tangutorum,of which 11 sites are same between hybrids and N.roborowskii,and 4 sites are same between hybrids and N.tangutorum.It is speculated that N.roborowskii is the female parent of hybrids and N.tangutorum is the male parent.The result of ITS sequence analysis revealed total 23 variable loci were detected from N.roborowskii and N.tangutorum,of which 10 sites are same between hybrids and N.roborowskii,and 7 sites are same between hybrids and N.tangutorum.In the remaining 6 loci,two different ITS sequence copies were found in each individual of hybrids.The results suggested that all intermediate individuals were the natural hybrids between N.roborowskii and N.tangutorum.In addition,we examined the sequence variation among the individuals of both parent species and hybrid swarms.The diversification of DNA fragments within the hybrid swarms probably resulted from the hybrid swarms experienced a longer period of multiplication.
