摘 要:目的 按表安排正交实验 ,优化渗漉法提取灵芝中抗肿瘤成分的条件 ,为灵芝的开发利用提供理论指导 .方法 乙醇浸泡后用渗漉法提取有效成分 ,以渗漉液对 He L a细胞的伤害程度作为衡量抗肿瘤成分相对含量的指标 .结果 灵芝子实体的醇提物在体外具有抑制 He L a细胞生长的作用 ;在影响渗漉条件的 4因素 (浸泡时间、漉速、用液量和操作温度 )中 ,温度影响最大 ,渗漉用液量影响最小 .结论 灵芝子实体中含有抑制 Hela细胞生长的成分 ,以浸泡 1 d,漉速 0 .5 m L· m in- 1 ,洗脱液用量 2 0 0 m L 和操作温度 60℃为条件能得到最大量的有效成分 .Aim To investigate the factors affecting the extraction of antitumor component from Ganoderma lucidum . Methods After being soaked in ethanol, the antitumor component was extracted by the method of percolation. The orthogonal design with table L 9(3 4) was taken. The relative content of antitumor component in the extract was estimated by its harmfulness to HeLa cell. Results The extract inhibits the growth of HeLa cell in vitro. Among the 4 factors affecting the percolation, the operating temperature was the most significant one associated with the harmfulness and the solvent volume was the least. Conclusion There were some components of inhibiting the growth of Hela cell in the fruiting body of Ganoderma lucidum. The percolating conditions: the soaking time of 1 d, the percolating rate of 0 5 mL·min -1 , the solvent volume of 200 mL and the temperature at 60 ℃, can ensure obtaining the best result of extraction.
