摘  要:目的:观察转化生长因子-β(TGF-β)和表皮生长因子(EGF)对人分泌中期子宫内膜间质细胞(ESC)基质金属蛋白酶-9(MMP-9)的影响。方法:体外培养ESC,分离、纯化后的ESC以1×105个/孔接种于96孔板,加入含10%胎牛血清的DMEM/F12培养液。24h后换无血清的DMEM/F12培养液,每孔200μL,分别加入TGF-β和EGF,终浓度均为2.5、5.0和10.0ng/mL,培养48h后通过ELISA法测培养液中MMP-9的含量。结果:与对照组比较,实验组随着TGF-β浓度的增加,ESC分泌的MMP-9明显下降(P<0.05,P<0.01);而随着EGF浓度的增加,ESC分泌的MMP-9则显著上升(P<0.05,P<0.01),且二者的变化具有剂量依赖性。结论:TGF-β和EGF可能通过调节人子宫内膜间质细胞MMP-9,而参与胚胎植入过程。Objective:To detect the expression of matrix metalloproteinases-9(MMP-9)in cultured human endometrial stromal cells(ESC)added different concentrations of transforming growth factor β(TGF-β)and epidermal growth factor(EGF).Methods:The isolated,purified ESC was planted to 1×105/well in 96-well plates,containing 10% FBS DMEM/F12 medium.After 24h culture,TGF-β and EGF were added to 200μl serum-free DMEM/F12 medium in each well and the final concentrations were 2.5ng/ml,5.0ng/ml and 10.0ng/ml,respectively.The content of MMP-9 was measured by ELISA after 48h culture.Results:Compared with the control group,the secretion of MMP-9 significantly decreased as the increasing concentration of TGF-β,while it was increase as the increasing concentrations of EGF,both showing a dose-dependent pattern.Conclusion:TGF-β and EGF may be involved in human embryonic implantation by regulating ESC secreting MMP-9.
