摘 要:目的观察全氟辛烷磺酸(PFOS)对SD大鼠的免疫损伤作用。方法健康成年SD大鼠24只,雌雄各半,随机等分为4组:溶剂对照组、低、中、高剂量PFOS染毒组;溶剂对照组进食2%Tween-80处理后的普通饲料,低、中、高PFOS染毒组分别进食5、25、125 mg/kg PFOS染毒饲料,连续染毒2个月。末次染毒24 h后,眼眶静脉丛取血后处死大鼠,测定各组大鼠的免疫功能指标的变化。结果中、高剂量PFOS染毒组脾脏、胸腺脏器系数比溶剂对照组降低(P<0.05);中、高剂量PFOS染毒组抗体形成细胞数、T淋巴细胞增殖功能、NK细胞的活性,及CD4+细胞、CD8+细胞、CD4+/CD8+比值及IFN-γ的分泌水平均低于溶剂对照组,差异均有显著性(P<0.05),并呈剂量—效应关系。与溶剂对照组比较,高剂量PFOS可降低大鼠对细胞的吞噬百分率和吞噬指数(P<0.05);脾脏组织出现不同程度的脾淋巴小结变小,边缘区变薄,白髓体积相对萎缩等病理损害,并随PFOS染毒浓度增加,病理损害越明显。结论全氟辛烷磺酸有致SD大鼠免疫功能损伤作用。Objective To observe the immunologic injury effection of Perfluorooctane Sulfonate(PFOS)on SD rats.Methods The 24 healthy SD rats(male and female in half)were divided into 4 groups:control group,low,medium and high doses of PFOS exposure group;control group was given fodder with 2% Tween-80;the experimental groups were given the treatment fodder with dose 5,25,125 mg/kg PFOS respectively for 2 months continuously(once a day);rats were killed in 24 hours after the last gastric perfusion,the blood from the orbital venous plexus was collected,then serum was prepared to determinate the immune function of various experimental groups.Results The spleen and thymus coefficient in the middle and high dose PFOS group were lower than those in the control group(P〈0.05),and there were dose-response relationships;All of the antibody-forming cells,the lymphocyte proliferation,CD4+T cells and CD4+ / CD8+,the NK cell activity and the level of IFN-γ in the middle and high dose PFOS groups were lower than those in the control group and had significant differences(P 0.05).Compared with resolver control group,the high dose PFOS could reduce SD rat's cell's swallow percentage and the phagocytic(P〈0.05);Spleen tissue appeared varying degrees changing:the spleen lymph nodules became smaller,marginal zone became thin,white cord volume was related to shrink and other pathological lesion,and all of these pathological lesions were obviously with the increase of the concentration of PFOS exposure.Conclusion PFOS could induce the immune toxicity injury of SD rats.
