无或低血清条件下高压培养促进平滑肌细胞和内皮细胞生长

摘  要:目的观察高压培养对无或低血清条件下平滑肌细胞和内皮细胞生长的影响。方法用无或0.5%血清培养基,在高于1个标准大气压(标准大气压设定为0 mmHg)30-180 mmHg压力下培养人脐静脉内皮细胞和大鼠血管平滑肌细胞。用台盼蓝染色细胞计数法和MTT评价细胞的生长情况。结果在低血清条件下,高压培养的血管内皮细胞生长速度明显快于标准大气压培养的血管内皮细胞,细胞计数和MTT结果显示高压培养比大气压培养细胞生长增加约30%。无血清大气压培养平滑肌细胞2天后,未见明显的活细胞,而高压培养大鼠平滑肌细胞仍能保持生长。结论无或低血清条件下,高压培养能保持血管平滑肌和血管内皮细胞的生长。Objective To investigate the effect of high pressure culture on smooth muscle cells(SMCs) and endothelial cells(ECs)in the condition of no or low serum.Methods In culture medium containing no or 0.5% serum,Human umbilical vein endothelial cells and Rat vascular smooth muscle cells were culture under one more standard atmospheric pressure(standard atmospheric pressure is set as 0 mmHg) from 30 to 180 mmHg.Trypan blue cell counting and MTT assay were performed to evaluate cells growth.Results Cell counting and MTT assay results showed high pressure culture increased about 30% of cells growth compared to standard atmospheric pressure culture in endothelial cells.Under the condition of no serum for two days,standard atmospheric pressure culture resulted in most of rat smooth muscle cells apoptosis,but high pressure culture still maintained cells growth.Conclusion High pressure culture increases the growth of vascular SMCs and ECs with no or low serum.
