摘  要:目的:观察连续单一应激(SPS)前阻断大鼠5-羟色胺1A(5-HTlA)受体下丘脑促肾上腺皮质激素释放因子(CRF)表达的改变,初步探讨5-HTlA受体活性与CRF表达的关系。方法:将健康雄性Wistar大鼠分为对照组、模型组和阻断组,模型组和阻断组给予SPS。阻断组在给予SPS应激前使用5-HTlA受体阻断剂WAYl00635进行干预。采用免疫组织化学和逆转录聚合酶链式反应(Rrr_PcR)方法检测下丘脑CRF蛋白和mRNA的表达。结果:对照组、模型组和阻断组免疫阳性细胞吸光度分别为6.94±0.61、22.36±1.55和16.86±1.65,mRNA相对表达分别为0.72±0.07、2.08±0.06和1.16±0.1t,模型组高于对照组,阻断组低于模型组。结论:5-HTlA受体活性与下丘脑CRF表达有关。XTo investigate the relationship between the expression of corticotropin-releasing factor (CRF) in hypothalams and the activity of 5-HT1A receptor in single-prolonged stress (SPS) rats. Methods: Adult male Wistar rats were randomly divided into control, model and blockade groups. The rats of the model group and the blockade group were exposed to SPS, and those in the blockade group were prior treated with WAY100635 (the 5-HT1A receptor antagonist). The expression of CRF was detected using immunohistochemistry and reverse transcription-polymerase chain reaction (RT- PCR). Results: The integral optical density of the model group (22.36±1.55) was significantly higher than that of the control group (6.94±0. 61), while that was lower in the blockade group (16.86± 0.07), and that in the blockade group (1.16 ±0. 1) was significantly decreased compared with the model group. Conclusion: SPS increased the expression of CRF, and the activity of 5-HTIA receptor was invloved the expression of CRF in hypothalamus in SPS rats.
