热稳定CHOD-CE-POD三合酶测定血清胆固醇试剂的研制与应用

摘　　要：用海藻糖作生化工具酶的常温干燥稳定剂，研制出热稳定良好的CHOD-CE-POD三合酶试剂，用于血清胆固醇的测定。此酶试剂热稳定性高，分别在70℃、55℃、37℃三种温度下放40d，150d、210d后再测酶活，三种酶的活性均保持在90％以上。酶试剂的线性范围为0～15．67mmol／L，批内CV＜1．5％，日间CV＜3．1％，回收率97．8％～100．5％，与中生公司酶法试剂盒（X）相关显著，r＝0．998，Y＝1．0206X－0．0162。An extraordinary stable CHOD-CE-POD mixed enzyme reagent was provided for serum cholesterol analysis. It based on the trehalose's enzyme protection characteristic. This enzyme reagent has high heat stabili-ty. After the enzyme reagent was stored at 37℃ for 210 days,at 55℃ for 150 days and at 70℃ for 40 days,it's activity was greater than 90%. The result was accurate and the repetition was good. The linearity was observed between 0~15. 67 mmol/L. Within-run CV was < 1. 5%,between Day CV was < 3. 0%. The rate of recovery was 97. 8% ~ 100. 5%. The result estimated by the CHOD- CE -POD enzyme reagent correlated closely with Changcheng's Cholesterol assay enzyme kit,and the correlative coefficient was r= 0. 998.
