摘 要:基于溶菌酶催化水解壳聚糖的产物氨基葡萄糖,与金溶胶-鲁米诺可产生很强的化学发光,建立了流动注射化学发光测定溶菌酶含量的方法。对水解和化学发光反应条件进行了考察。结果表明,pH 4.0、50℃水解5 h为最佳水解条件。在金胶、鲁米诺及NaOH的浓度分别为0.05,0.05和10 mmol/L条件下,化学发光检测效果最好。在优化条件下,溶菌酶浓度在5~800μg/L范围内与化学发光强度有良好的线性关系,方法的检出限为4μg/L。本方法可用于蛋清中溶菌酶含量的检测。Basing on the fact that glucosamine which is hydrolysate of chitosan with lysozyme can react with luminol-gold nanoparticles giving birth to chemiluminescence,a flow injection chemiluminescence(CL) method was proposed for the detection of lysozyme.The hydrolysis condition and the CL condition were studied.It was found that the optimized experimental conditions of hydrolysis were pH 4.0,50 ℃ and hydrolysis for 5 h.When the concentrations of gold colloids,luminol and NaOH were 0.05,0.05 and 10 mmol/L respectively,the optimized results of CL measurements were obtained.Under the conditions,a linear relationship was obtained between the CL intensity and the concentrations of lysozyme in the range of 5 to 800 μg/L,with the detection limit of 4 μg/L.This method has been successfully applied to the determination of lysozyme in egg white.
