摘  要:目的:观察肾性高血压(RH)时,血浆及血管组织中内皮素(endothelin, ET)及基础NO的动态变化,探讨其在肾性高血压发生发展中的作用。方法:将体重为160~180 g 雄性Wistar大鼠随机分为肾性高血压组及假手术组,术后饲养2,4,6 周后测定大鼠血压的变化、血浆及主动脉中ET 含量、主动脉基础一氧化氮(nitricoxide, NO)释放量。结果:RH组术后2周时血压即已升高,4、6 周时显著升高。RH组2,4,6周基础NO生成量显著低于同期C组,而血浆及主动脉中ET含量均显著高于同期C组。结论:肾性高血压时,血管内皮细胞功能紊乱,维持血管正常张力的基础NO生成减少。Objective:To observe the dynamic change of endothelin(ET) in plasma and aortic tissue, basal NO release from aortic endothelium in renal hypertensive rats orderto explore the roles of them in the initiation and maintenance of renal hypertension. Methods: Male Wistar rats weighing 160~180 g were divided into two groups: renal hypertension (RH) group and control(c) group and fed for 2,4,6 weeks respectively. Blood pressure, basal NO, plasma and aorta ET were measured by inserting a cannula into the carotid artery using bioassay and radioimmunoassay respectively. Results: The blood pressure increased in 2 weeks and markedly increased in 4,6 weeks in RH rats. Basal NO release from endothelial cells of 2,4 6 weeks RH rats was reduced. Meanwhile, the contents of ET in plasma and aortic tissue were markedly increased. Conclusions:The vascular endothelial cells function of renal hypertension rats was disturbed. The balance of vascular regulating system, basal NO (relaxation) and ET (contraction),was impaired in RH rats. This might be involved in the initiation and maintenance of renal hypertension.
