摘  要:目的研究大鼠生长发育期氯化镧(lanthanum chloride,LaCl3)暴露对大鼠大脑学习记忆能力和皮质即刻早期基因c-jun、egr1和Arc表达的影响。方法将40只Wistar孕鼠随机分为饮用0.25%、0.5%、1.0%LaCl3蒸馏水溶液的3个染毒组和饮用蒸馏水的对照组。染毒组仔鼠断乳后继续分别自行饮用0.25%、0.5%和1.0%LaCl3蒸馏水溶液1个月。采用水迷宫试验测试仔鼠的学习记忆能力,采用电感耦合等离子体质谱仪测定仔鼠大脑皮质镧含量,采用RT-PCR法检测仔鼠大脑皮质c-jun、egr1和Arc mRNA表达,采用western blot法检测仔鼠大脑皮质c-jun、egr1和Arc蛋白的表达。结果水迷宫测试结果表明,0.25%、0.5%LaCl3组仔鼠错误次数和逃逸潜伏期明显高于对照组,1.0%LaCl3组仔鼠错误次数和逃逸潜伏期明显高于对照组和0.25%LaCl3组,且错误次数明显高于0.5%LaCl3组,差异均有统计学意义(均P<0.05)。镧在仔鼠大脑皮质中的蓄积量随着LaCl3染毒浓度的增加而增加。此外,0.25%、0.5%LaCl3组仔鼠大脑皮质c-jun和egr1 mRNA和蛋白表达水平显著低于对照组,1.0%LaCl3组仔鼠大脑皮质c-jun mRNA和蛋白表达水平显著低于对照组和0.25%LaCl3组,且egr1 mRNA和蛋白表达水平显著低于对照组和0.25%、0.5%LaCl3组,差异均有统计学意义(均P<0.05)。结论生长发育期LaCl3暴露可影响大鼠在水迷宫中的学习记忆能力,其机制可能与大脑皮质即早基因c-jun和egr1基因和蛋白表达降低有关。Objective To study the effects of lanthanum chloride (LaC13) on the ability of learning and memory and immediate early genes including c-jun, egrl and A rc expression in the cerebral cortex of rats during growth and development period. Methods Forty Wistar pregnant rats were randomly divided into three groups treated with LaCl3 at 0.25%, 0.5% and 1.0% LaC13 through drinking water and control group drinking distilled water. After weaning, the pups in LaC13 groups continued to he treated with LaCl3 at 0.25%, 0.5% and 1.0% through drinking water for one month. Learning and memory of pups were measured by water maze. Lanthanum contents in the cerebral cortex of pups were measured by inductively coupled plamsa mass spectrometry (ICP-MS). C-jun, egrl and Arc mRNA expression in the cerebral cortex of pups were detected by RT-PCR, and c-jun, egrl and Arc protein expression in the cerebral cortex of pups were measured by western blot. Results The results in the water maze test showed that wrong frequency and escape latency of pups in 0.25% and 0.50% LaCl3 groups were higher significantly than those in control group, these parameters of pups in 1.0% LaCl3 groups were higher significantly than those in control and 0.25% LaCl3 groups and wrong frequency of pups in 1.0% LaCl3 groups were also higher significantly than that in 0.5% LaCl3 group. The accumulation of lanthanum in the cerebral cortex of pups increased with the exposure of LaCl3 increasing. Besides, c-jun and egrl mRNA, protein expression levels in the cerebral cortex in 0.25% and 0.5% LaCl3 groups were significantly lower than those in control group. C-jun mRNA and protein expression levels of pups in 1.0% LaCl3 group were significantly lower than those in control and 0.25% LaCl3 groups, and egrl mRNA and protein expression levels of pups in 1.0% LaCl3 group were lower significantly than control, 0.25%, and 0.5% LaCl3 groups.
