摘 要:目的探讨钙和大豆异黄酮的联合作用对去卵巢大鼠骨生物力学特性的影响。方法将160只12月龄SD清洁级雌性大鼠随机分为4组,分别为假手术对照(蒸馏水)组、骨质疏松模型(蒸馏水)组、大豆异黄酮(10 mg/ml,染毒容量为3 ml/kg)组、雌激素(0.1 mg/ml,染毒容量为0.1 ml/kg)组,每组再按照加钙(150 mg/ml,染毒容量为1 ml/d)和不加钙(蒸馏水)分为两组,每组20只。连续染毒6个月。分别在染毒3和6个月时,测量股骨的6项生物力学指标(扭转刚度、柔韧系数、最大载荷、破坏扭转角、最大正应力、破坏能量吸收)并进行统计分析。结果染毒3个月,钙能显著提高大鼠股骨柔韧系数、最大载荷和破坏能量吸收的指标值;大豆异黄酮、钙与大豆异黄酮的交互作用能显著提高股骨扭转刚度、柔韧系数、最大载荷、最大正应力、破坏能量吸收5个生物力学指标值(对破坏扭转角影响不显著)。染毒6个月,钙、大豆异黄酮以及钙与大豆异黄酮的交互作用均能显著提高股骨6个生物力学指标的均值,与染毒3个月时相比,影响效果均显著增强(P<0.05)。结论钙与大豆异黄酮联合作用可以明显改善骨生物学性能。Objective To investigate the effect calcium and soybean isoflavone in combination on the biological mechanics in ovariectomized rats. Methods The tests were conducted through gavage. One hundred and sixty female SD rats were randomly divided into four groups, as sham operated group (distilled water), osteoporosis model group (distilled water), soybean isoflavone group ( 10 mg/ml, administration dosage 3 ml/kg)and estrogen group (0.1 mg/ml, administration dosage 0.1 ml/kg). Each group was divided into two subgroups as calcium ( 150 mg/ml, administration dosage 1 ml/d) and no-calcium subgroups (distilled water)with 20 rats in each subgroup, and further divided into two batches (the first batch was sacrificed three months later, the second one six months later). The femoral biological mechanics indices (torsional rigidity,the coefficient of flexility,mutilated torsion angle, maximum load,maximum normal stress and energy absorption for break)were measured. Results After three months of treatment,the calcium could markedly increase the coefficient of flexility,maximum load and energy absorption for break;The estrogen and the combination of calcium and estrogen could markedly increase the five biological mechanics indices (torsional rigidity, the coefficient of flexility, mutilated torsion angle, maximum load, maximum normal stress, energy absorption for break), there was no marked influence on mutilated torsion angle. After six months of treatment,calcium and estrogen in combination presented the best effect in the six biological mechanics indices. Conclusion The combination of calcium and soybean isoflavone can distinctly improve the biological properties of bone inrats.
