摘 要:目的探讨污水有机提取物对小鼠脾细胞的DNA损伤作用。方法于2004年春季(4月)和秋季(10月)采集某市污水灌溉地区水样。将1只健康8周龄清洁级雄性昆明小鼠处死,调整脾脏细胞悬液密度为2×106/ml,分别加入终浓度0(溶剂对照)、0.312 5、0.625、1.25 ml/L的污水有机物提取液,于37℃染毒2 h,并设阳性对照(0.2 mmol/L重铬酸钾)。采用非荧光染色彗星试验检测污水有机提取物对小鼠脾细胞的DNA损伤作用。结果与溶剂对照组比较,各剂量组春季和秋季污水有机提取物染毒小鼠脾细胞的拖尾率和彗星尾长均较高,差异有统计学意义(P<0.05)。随着污水有机提取物染毒剂量的升高,小鼠脾细胞拖尾率和彗星尾长均增加,两者之间呈剂量-反应关系(P<0.01)。结论污水中的有机物可诱发小鼠脾细胞发生DNA损伤。Objective To discuss the DNA damage effect of organic extracts from sewage on the mouse spleen cells.Methods The water samples were collected in the spring(April) and autumn(October) of 2004 in sewage irrigation area.The spleen cell suspension was prepared from a clean and healthy 8-week old male Kunming mice,the cell density was 2 ×106/ml.2 μl organic extracts of sewage[final concentration was 0(solvent control),1.25,0.625 and 0.312 5 ml/L respectively] were added to 2 ml of spleen cell suspension,for 2 h at 37 ℃,and the positive control group was treated with potassium dichromate(0.2 mmol/L).The DNA damage effect was detected by using un-fluorescent staining comet assay.Results Compared with the solvent control group,significant differences were observed in the tailed-cells rates and tail's length of the cells in the treated groups with a dose dependent manner.Conclusion Organic extracts of sewage may induce DNA damage in mouse spleen cells in vitro.
