摘 要:目的产丙酮酸棒状杆菌是近年来发现的细菌新种,其与痤疮丙酸杆菌一样属需脂杆菌,但有无与痤疮丙酸杆菌类似的免疫调节作用尚不清楚。本文以痤疮丙酸杆菌为对照,以在固有和适应性免疫应答中发挥重要调节作用的树突状细胞为靶向,研究了产丙酮酸棒状杆菌对树突状细胞成熟标志表达和细胞因子产生等方面的影响,为该菌作为免疫调节剂进一步开发利用提供依据。方法加热灭活的产丙酮酸棒状杆菌或痤疮丙酸杆菌刺激树突状细胞DC2.4后,ELISA法检测其产生相关细胞因子的变化;MTT法检测细胞的增殖情况;流式细胞仪分析细胞表面MHCⅡ、CD40等表达水平。结果两种细菌刺激后,DC2.4表达IL-6和IL-12的水平均有增加,且随刺激的菌液浓度的增加而升高,这种刺激还明显增强DC2.4的增殖能力。此外,刺激后细胞表面的MHCⅡ、CD40/80/86表达也呈现不同程度的上调,尤以MHCⅡ为著。结论产丙酮酸棒状杆菌对DC2.4细胞增殖、成熟和相关细胞因子的表达均有明显的影响,这种影响将有助于促进DC的抗原提呈功能。Corynebacterium Pyruviciproducens(CP) is a newly discovered bacteria,like Propionibacterium acnes(PA),belonging to the adipo-bacillus.However,we still do not know whether CP has the same immunoregulation with PA or not.Referring to PA,we studied the immunomodulatory effect of CP on mouse dendritic cells,which play an important role in inherent immunity and adaptive immunity.All of that provides researches on immune adjuvant of CP with a theoretical basis.After DC2.4 cells cocultured with heating-killed PA or CP,we detected some related cytokines and determined the cell proliferation by MTT,and checked the expression of MHCⅡ CD40 and other surface molecules by FACS.The data showed that the expression of IL-6/12 increased depending on the concentration of PA/CP,and the ability of DC2.4 cell proliferation was markedly enhanced by PA/CP stimulation.Furthmore,DC2.4 cells showed higher levels of MHCⅡ and CD40/80/86 after stimulation than unstimulated,especially MHCⅡ.On the whole,CP can promote the production of IL-6/12 and the proliferation of DC2.4 and induces the maturation of DC,which would facilitate the antigen presentation of DC.
