摘  要:目的 了解产青霉素酶淋病奈瑟菌(PPNG) 株的耐药性特征、耐药与质粒携带类型之间的相互关系以及追踪耐药菌株的来源。方法 对本地区1996 年5 ~7 月分离的48 株PPNG 标本进行了分析。用琼脂扩散法和琼脂稀释法测定标本对11 种抗生素的敏感性。碱变性加溶菌酶法对标本进行质粒抽提。电洗脱法回收5-3kb 和7-4kb 质粒,用Bam HⅠ单切酶和Bam HⅠ+ Pst Ⅰ、Bam HⅠ+ BgLⅠ双切酶酶切分析。结果 48 株PPNG 株对青霉素G、四环素和氟哌酸耐药率较高,分别为100 % 、93-7 % 和89-58 % ,对其余抗生素较敏感。48 株PPNG 菌株中47 株被检出耐药性质粒,但耐药性质粒酶切片段多少不一,具有明显的多态性。结论 耐药性质粒酶切分析为追踪耐药性质粒来源及耐药菌株的流行趋势提供了流行病学信息。Objective To investigate the characteristics of drug tolerance,the correlation of antibiotic resistance with plasmid carrying type,and to pursue the source of antibiotic resistant strains,48 PPNG strains were determined for susceptibility of 11 antibiotics,plasmid profiles,and tolerance plasmids analysed by restriction endonuclease. Methods Susceptibilities to 11 antibiotics had been done with agar diffusion method and agar dilution method.Plasmids were extracted by alkali with lysozyme.The plasmids 5.3kb and 7.4kb were removed from agarose gel by electroelution method.And meanwhile plasmids DNA were digested by BamHⅠ、BamHⅠ+PstⅠ and BamHⅠ+BgLⅠ. Results The resistance rate of 48 PPNG strains to Penicillin G,Tetracycline,and Norfloxacin was 100%,93.76%, and 89.58%,respectively,but to the rest antibiotics were sensitive comparatively.Forty seven strains possessing tolerance plasmids were found in 48 PPNG strains.Analysis of the plasmid fingerprinting there were polymorphic in 5.3kb and in 7.4kb. Conclusions The analysis of restriction endonuclease can pursue the source of tolerance plasmids,and at the same time get the epidemiologic information about the antibiotic resistance strains′epidemic trend.
