摘  要:目的:研究吴茱萸提取物(EV)对野百合碱(MCT)所致大鼠右室肥大及钙调神经磷酸酶的抑制作用。方法:将60只雄性Wistar大鼠均分为正常对照、右室肥大模型、硝苯地平(20mg·kg-1)和EV低、中、高剂量(50、100、200mg·kg-1)组。连续ig给药18d,测定大鼠右室肥大指数、右室相对质量和肺重/体重,实时定量PCR检测心肌组织钙调神经磷酸酶(CaN)mRNA的表达,免疫组化染色测定CaN催化亚基(CnA)的蛋白表达。结果:与模型组比较,EV50、100、200mg·kg-1使右室肥大指数、右室相对质量和肺重/体重显著降低(P<0.01或P<0.05),CaNmRNA和CnA的蛋白表达显著降低(P<0.01)。结论:EV能明显改善MCT引起的大鼠右心室肥大,其机制可能与其抑制CaN信号转导通路有关。OBJECTIVE:To observe the inhibitory effect of the extracts of Euodiae Fructus(EV) and monocrotaline(MCT) on right ventricular hypertrophy(RVH) and calcineurin of rats.METHODS:60 male Wistar rats were randomly divided into 6 groups:normal control group,right ventricular hypertrophy model group,nifedipine(20 mg·kg-1)group,Euodiae Fructus.rutaecarpa low-dose,medium-dose and high dose groups(50,100,200 mg·kg-1).The rats were given drugs intragastrically for 18 days.RVH index,RV weigh/body weigh(RVW/BW),lung weight/ body weigh(LW/ BW) of rats were measured.The expressions of calcineurin(CaN) mRNA and CnA protein were separately determined by real-time RT-PCR and immunohistochemistry.RESULTS:Compared with model group,Euodiae Fructus(50,100,200 mg·kg-1) could significantly decrease RVH index,RVW/BW and LW/ BW of rats(P0.01 or P0.05),at the same time the expressions of CaN mRNA and CnA protein were significantly suppressed by Euodiae Fructus(P0.01).CONCLUSION:Euodiae Fructus can significantly attenuate the right ventricular hypertrophy induced by MCT in rats,its mechanism may be associated with inhibition effect of it on CaN signal transduction pathway.
