摘  要:目的对哈尔滨市污水处理厂原水中隐孢子虫进行分子鉴定。方法提取污水处理厂原水中的隐孢子虫基因组DNA,对其18S rRNA基因进行巢式PCR扩增,通过序列分析确定隐孢子虫虫种,并与GenBank的参考序列进行同源性分析。结果从哈尔滨市污水处理厂原水中分离鉴定出安氏隐孢子虫,该虫株18S rRNA基因序列与文献报道的安氏隐孢子虫序列(EU926592)同源性为100%。结论哈尔滨市污水处理厂原水中存在安氏隐孢子虫,对当地畜牧业的发展构成严重威胁,但对当地人群危害较小。Objective To molecularly identify a Cryptosporidium isolate from raw urban wastewater at a wastewater treatment plant in Harbin.Methods Genomic DNA of Cryptosporidium was extracted.The Cryptosporidium species was identified by nested PCR amplification of the 18S ribosomal RNA gene and sequenced.The homology of the gene was analyzed while referring to nucleotide sequences downloaded from GenBank.Results C.andersoni was identified in raw urban wastewater at a wastewater treatment plant in Harbin.The obtained sequence at the 18S ribosomal RNA locus had 100% homology with a previously reported sequence of C.andersoni(EU926592).Conclusion C.andersoni was found in raw urban wastewater at a wastewater treatment plant in Harbin.This parasite poses a threat to the growth of the local livestock industry but is less of a threat to the local population.
