摘 要:目的:建立并优化大鼠在体胃缺血后处理模型,并初步探讨缺血后处理对胃黏膜损伤的抗氧化机制。方法:78只大鼠分为13组,每组6只。采用单纯胃缺血-再灌注(GI-R)损伤组和假手术组作对照,通过检测胃黏膜损伤指数,观察缺血后处理时短暂的再灌注(R)-缺血(I)持续时间(R和I分别均为10、20、30和40s组)、R-I循环次数(R20s/I20s反复1、3、5和10次组)以及长时间再灌注开始后给予后处理的时间(R为1、2和4min组),对胃黏膜再灌注损伤的影响,并测定胃黏膜组织丙二醛(MDA)含量与超氧化物歧化酶(SOD)活性的变化。结果:假手术组、GI-R组及4个不同持续时间组胃黏膜损伤指数比较,差异有统计学意义(F=16.574,P<0.001);假手术组、GI-R组及4个不同循环次数组胃黏膜损伤指数比较,差异有统计学意义(F=34.815,P<0.001);假手术组、GI-R组及3个不同开始时间组胃黏膜损伤指数比较,差异有统计学意义(F=48.499,P<0.001)。各组大鼠胃黏膜MDA含量(F=27.250、31.450和27.520,P均<0.001)和SOD活性(F=72.662、57.220和50.650,P均<0.001)比较,差异均有统计学意义。结论:成功构建并优化了大鼠胃缺血后处理模型。Aim: To establish and optimize the gastric ischemic postconditioning(GI-postc) model in rats,and preliminarily investigate the antioxidation mechanism of GI-postc.Methods:A total of 78 rats were randomly divided into 13 groups.Sham group and GI-R group were used as control.The experimental groups included four different R and I time groups (10,20,30,and 40 s R/I),four different cycle groups (1,3,5,and 10 cycles),and three different beginning time groups(1,2,and 4 min).Gastric mucosal injury index was detected.The content of malondialdehyde (MDA) and the activity of superoxidase dismutase (SOD) in gastric mucosa were detected.Results: GI-postc could significantly reduce gastric mucosal injury induced by I/R.There were significant differences among sham group,GI-R group,and four different R and I time groups(F=16.574,P0.001),among sham group,GI-R group,and four different cycle groups(F=34.815,P0.001),among sham group,GI-R group,and three different beginning time groups (F=48.499,P0.001).There were significant differences on MDA(F=27.250,31.450,and 27.520,P0.001) and SOD (F=72.662,57.220,and 50.650,P0.001) among these groups.Conclusion:The GI-postc model in rats has been established and optimized successfully.
