吗啡依赖和戒断大鼠某些脑核团中β-内啡肽含量的变化

摘　　要：为探讨阿片依赖和戒断的神经生物学机制,本研究观察了吗啡依赖和戒断对大鼠脑内与奖赏效应及戒断症状有关核团中β内啡肽(βEP) 含量的影响。结果:(1)吗啡依赖大鼠伏隔核(Acc) 、中脑导水管周围灰质(PAG) 、蓝斑核(LC) 和下丘脑弓状核(ARH) 的βEP含量显著低于正常对照组( P < 0 .05 或0 .01) 。(2) 吗啡戒断大鼠Acc 和ARH 的βEP含量与依赖组比较无显著差异,仍显著低于正常对照组( P< 0 .05 或0 .01) ;而VTA( 腹侧被盖区) 、PAG 和LC 的βEP 含量较依赖组显著升高( P< 0 .05) ,与正常对照组比较无显著差异。结论:吗啡依赖和戒断时脑内与奖赏效应及戒断症状有关的一些核团中β内啡肽含量发生了改变。To investigate neurobiological mechanisms of opiate dependence and abstinence, the contents of β endorphine in brain areas related with rewarding and withdrawal syndrome in rats were determined in morphine dependent and abstinent rats by radioimmunoassay. Results:(1)In morphine dependent group,the β EP contents in nucleus accumbens(Acc),periaqueductal gray(PAG),locus coeruleus(LC) and nucleus arcuatus hypothalami(ARH) were significantly lower than that in control group (P<0.05 or 0.01).(2)In morphine abstinent group,the β EP contents in Acc and ARH were significantly lower than that in control group (P<0.05 or 0.01),and were not significantly different from that in morphine dependent group;however,the β EP contents in VTA, PAG and LC were not different from that in control group, and were significantly higher than that of morphine dependent group(P<0.05 or 0.01).Conclusion:There is a change of the β endorphin contents in brain nuclei associated with rewarding and withdrawal syndromes during the development of morphine dependence and abstinence.
