摘  要:目的基于5-HT能神经系统,探究四逆散冻干粉改善睡眠的作用机制。方法分别观察四逆散冻干粉与5-HT前体物质5-HTP、5-HT拮抗剂PCPA、5-HT1A受体拮抗剂p-MPPI和5-HT2受体拮抗剂Ritanserin联用,对戊巴比妥钠所致小鼠睡眠时间的影响。结果阈下剂量四逆散冻干粉联合5-HTP可协同延长戊巴比妥钠所致的小鼠睡眠时间(P<0.05);阈剂量四逆散冻干粉对PCPA化小鼠睡眠时间缩短有对抗作用,PCPA亦可对抗四逆散冻干粉的催眠作用;5-HT1A受体拮抗剂可明显拮抗四逆散冻干粉的催眠作用(P<0.01);四逆散冻干粉联合5-HT2受体拮抗剂,与单独给予四逆散冻干粉没有显著性差异。结论四逆散改善睡眠作用主要依赖于5-HT的存在,且延长戊巴比妥钠所致小鼠睡眠时间与5-HT1A受体有关,与5-HT2受体无关。Objective To elucidate the mechanism of the effect of Sinisan lyophilized powder on promoting sleep based on the nervous system of 5-HT.Methods Effects of SINISAN combined with 5-HT precursors 5-HTP,5-HT antagonist PCPA,5-HT1A receptor antagonist p-MPPI and 5-HT2 receptor antagonist ritanserin on sleep time in mice pretreated with pentobarbital were investigated.Results Subthreshold doses of Sinisan lyophilized powder combined with 5-HTP significantly prolonged the time of sodium pentobarbital-induced sleep in mice(P0.05);Subcutaneous injection of PCPA significantly shortened the time of sodium pentobarbital-induced sleep in mice,meanwhile,the threshold dose of Sinisan obviously antagonized the effect of PCPA.Moreover,p-MPPI obviously antagonized of the effect of Sinisan on prolonging the effect of sodium pentobarbital-induced sleep in mice,when treated with Sinisan and p-MPPI or ritanserin,there was no significant difference between merger ritanserin or given alone Sinisan lyophilized powder.Conclusion The existence of Sinisan prolongs sodium pentobarbital-induced sleep time in mice has nothing to do with the 5-HT2 receptor but the 5-HT1A receptor.
