摘 要:目的观察感染尖吻蝮蛇舌形虫小鼠体内特异性抗体和循环抗原动态变化。方法从感染尖吻蝮蛇舌形虫的五步蛇体内收集成虫,分离尖吻蝮蛇舌形虫的虫卵感染小鼠,从感染后1 w2、w、3~22 w收集小鼠血清,分别用ELISA法和dot-ELISA法观察不同时间小鼠体内尖吻蝮蛇舌形虫特异性抗体和循环抗原的动态变化。结果感染尖吻蝮蛇舌形虫的小鼠,其体内特异性抗体从第8 w开始上升,12 w达到高峰,第16 w开始下降并一直维持同一水平。同时,对小鼠产生的特异性抗体进行分型,其最早出现为IgM,16 w以后被IgG1所替代。感染尖吻蝮蛇舌形虫的小鼠血清用dot-ELISA法检测其循环抗原出现的时间为第1 w,到第3 w时循环抗原检出稀释度在1∶8~1∶128之间,到第8 w,最高稀释度可达到1∶256,并一直维持,第11 w以后逐渐下降。结论 1.小鼠感染尖吻蝮蛇舌形虫虫卵后特异性抗体(Ab)在感染第8 w开始上升,抗体最高滴度维持时间为第12~15 w。2.感染尖吻蝮蛇舌形虫虫卵的小鼠其血清中产生的特异性抗体最早出现为IgM,以后被IgG1所替代。3.小鼠在感染尖吻蝮蛇舌形虫虫卵后的1 w就可在其血清中检测到循环抗原(CAg),预测这段时间检测循环抗原具有早期诊断参考价值。In this study, adult A. agkistrodontis were collected from hundred-pace snakes, and then the eggs were sepa- rated to feed mice. In the next step, when the infection model was established, blood serum of infected mice were collected after one, two and three weeks, respectively. Furthermore, EIASA and dot-ELISA were both used to detect the dynamic observation of specific antibodies and circulating antigens. The specific antibodies increased from 8th week, reached the top at 12th week, decreased from 16th week, and then maintain the same l.evel constantly. Meanwhile, the specific antibodies were typing. It is evidedent that IgM antibody appeared earliest. However, the former was substitute by IgG1 after 16th week. Moreover, the circulating antigens have been detected in the first week by dot-ELISA. Then, the dilute strength between 1 : 8- 1 : 128 were founded in 3rd week. The highest dilution strength with 1 : 256 appeared at 8th week, which would maintain before 11th week and decreased gradually, which will be valuable for early diagnosis of circulating antigens.
