摘  要:目的探讨不同毒力疟原虫感染早期根治性治疗对再感染细胞免疫应答的影响。方法用2种毒力不同约氏疟原虫感染DBA/2小鼠,感染后3 d进行根治性治疗,并于感染后90 d进行再感染。吉姆萨薄血膜染色法计数红细胞感染率,流式细胞术检测再感染前后不同时间点脾T细胞中活化性T细胞百分率,ELISA检测脾细胞培养上清中细胞因子水平。结果两组根治性治疗小鼠再感染后均出现了短暂的低水平虫体血症;活化性T细胞百分率、IFN-γ、TNF-αI、L-10和IL-4分别于再感染后第1 d、3 d和5 d出现有意义的升高;但在每一相同检测时间点,两组小鼠的虫体血症、活化性T细胞百分率、Th1和Th2型细胞因子水平均没有显著差异。结论不同毒力疟原虫感染早期根治性治疗后,宿主在同种疟原虫再感染时可产生模式和强度相似的细胞免疫应答。To investigate the effect of radical treatment in early phase of primary infection with different toxicity plasmo- dium on cellular immune response to homologous reinfection, DBA/2 mice were infected respectively with Py17XL and Pyl7XNL parasited erythrocytes, and radically treated on day 3 after infection. The mice were reinfected respectively with Py17XL and Pyl7XNL on day 90 after primary infection, then the level of parasitemia was observed by mean of the Giemsa staining of thin blood smears, and applying flow cytometry to quantitively analyze the percentage of actived T cells in spleen T cells on days 0, 1,3 and 5 post-reinfection, respectively, the levels of IFN-γ, TNF-α, IL-4 and IL-10 in spleen cell culture supernatant were also measured by ELISA at the same time. Results showed that parasitemia in two groups of radically treated mice post-reinfection was all transient and extremely low, and percentage of actived T cells, levels of IFN-γ, TNF-α, IL-10 and IL-4 began to rise from day 1 to day 5 after reinfection, respectively, in each testing time point, however, the levels of parasitemia and percentage of actived T cells as well as levels of these cytokines from two groups of mice were not significantly different. These results suggest that host radically treated in early phase of primary infection with different toxicity malarial para site may produce a similar pattern and intensity of cellular immune responses to homologous reinfection.
