摘 要:目的探讨大鼠脊髓损伤后应用蛇毒神经生长因子对Caspase-3表达的影响。方法将55只成年SD雄性大鼠随机分为蛇毒神经生长因子组(A组)、生理盐水组(B组)、假手术组(C组),按改良Allen打击法建立大鼠脊髓不完全损伤模型,通过动物神经运动功能BBB评分评价神经损伤程度及神经功能恢复情况;脊髓损伤后不同时间点(6 h、1 d、3 d、7 d、14 d)取材,HE染色观察损伤脊髓组织病理变化,免疫组化染色检测Caspase-3阳性细胞的表达,来比较分析两组的差异性。结果 HE染色镜检发现脊髓组织病理学改变A组明显轻于B组;BBB评分A组相对B明显提高,差异有显著性意义(P<0.05)。A组和B组均发现凋亡细胞及Caspase-3表达,神经细胞凋亡指数及Caspase-3表达均为B组>A组(P<0.05)。结论蛇毒神经生长因子能抑制大鼠脊髓损伤后Caspase-3表达及细胞凋亡,能改善脊髓损伤后的功能表现。Objective To investigate the effect of Snake Nerve Growth Factor (NGF)on Caspase-3 expression after spinal cord injury (SCI) in rats. Methods Fifty-five adult SD rats were randomly divided into Snake Nerve Growth Factor (group A),normal saline (group B), sham group (group C),the SCI model was induced according to improved allen's method,the neurofunctional recovery was observed according to BBB score, after operation, the rats was sacrificed in different time (6 h, 1 d, 3 d, 7 d, 14 d), and the injuried spinal tissue specimens were stained with hematoxylin and eosin (HE),immunohistochemistry examines the Caspase-3 masculine cell rate change to carry on the analysis. Results The histopathologic changes in spinal cord tissues in group B was worse than that in group A. The BBB score in A group was significantly differnce with normal saline group B(P〈0.05). The expression of Caspase-3, p53 and apoptotic cells was observed in two groups,and the apoptosis index and the expression of Caspase-3,p53 in group B were higher than that in group A(P〈0.05). Conclusion Snake Nerve Growth Fctors could inhibit the expression of Caspase-3 and apoptosis after SCI in rats.
