PHA复方制剂对鸡免疫功能及相关细胞因子的调节作用

摘　　要：试验旨在探讨由植物血凝素(PHA)、黄芪多糖为主药组成的植物血凝素复方制剂对新城疫疫苗免疫鸡的免疫功能和Th1/Th2型细胞因子的调节作用。研究测定了不同处理组中鸡脾脏淋巴细胞培养液IFN-γ、IL-10的浓度,并将复方制剂按体重0.05、0.2、0.3 g.kg-1三个剂量灌服鸡,对不同日龄Lasota疫苗免疫鸡免疫脏器指数及血清中IFN-γ、IL-10的浓度进行测定。结果表明,在体外,PHA在调节Th1型IFN-γ细胞因子时发挥主要作用,黄芪多糖在调节Th2型IL-10时发挥主要作用;在体内,复方制剂中剂量组可以显著性提高胸腺、脾脏、法氏囊的脏器指数,并在免疫鸡外周血中有短时促进Th2型IL-10的产生,降低Th1型细胞因子IFN-γ的产生。结果显示,植物血凝素复方制剂对新城疫免疫鸡有免疫增强作用,作用机理与调节Th1/Th2细胞因子有关。The immune function influence of ND Immune chicken and the mediation role to Th1/Th2 about the preparation to be explored,the compound preparation was composed with PHA and astragalus polysaccharides.This study detemined immune organs index and the concentrations of IFN-γand IL-10 in different chicken spleen lymphocytes broth and in different days Lasota vaccine immune chicken serum before that the compound preparation were gavaged at doses of 0.05,0.2,0.3 g·kg-1.The results showed that the compound preparation could relate cytokines coordinated.The PHA played a major role in regulating IL-10 in Th2 behalf and astragalus polysaccharides played a major role in regulating IFN-γ in Th1 behalf in vitro.In vivo,the compound preparations could significantly improve the thymus and spleen and bursa of Fabricius indexes.The compound preparations had short-time role of promoting IL-10 in Th2 behalf and turning down IFN-γ in Th1 behalf in immune chicken＇s peripheral blood.The compound preparations to ND immunity chickens have immuno-enhancing which had relevants to Th1/Th2.
