摘  要:利用5 L气球型气升式生物反应器研究蔗糖和氧化锗浓度对不定根生长和人参皂苷合成的影响,结果表明,当MS+IBA 5 mg/L培养基中加入蔗糖50 g/L时,每个反应器可收获不定根34.5 g(干重),皂苷生产量达到61.1 mg/L,显著好于蔗糖30和70 g/L处理;培养基中加入氧化锗30或60μmol时,促进不定根鲜重的增加,干重和增殖率在60μmol时最好;不定根生长30 d后将60μmol的氧化锗加入培养基时,促进皂苷的合成,氧化锗处理12 d后皂苷含量由5.66 mg/g DW增加到10.8 mg/g DW.对反应器培养不定根生长、皂苷合成及培养基变化动态进行研究结果,不定根生物量(鲜重和干重)和皂苷生产量随着培养时间的延长而增加,培养30 d时生物量达到最高值,但培养时期继续延长时生物量则不再增加,而皂苷生产量却在40 d时最高。因此,为获得更多的皂苷,反应器不定根培养时期以40 d为最佳。培养基中糖浓度的变化与生物量的变化呈负相关;pH在培养初期呈下降趋势,以后随培养时间逐渐上升,培养结束时接近培养初期值.The effect of sucrose and germanium on adventitious root growth and ginsenoside synthesis of in Panax ginseng were carried out, the results showed that the medium of MSq-5 mg/L IBA supplemented with 50 g/L sucrose was significantly better than the concentrations of 30 g/L and 70 g/L sucrose , dry weight and ginsenoside productivity reached 34.5 g and 61.1 mg/L, respectively. 60μmol GeO2 in the cul- ture medium promoted adventitious root biomass, especially dry weigth, the growth coefficient was 24.7. Ginsenoside content did not change when GeO2 was supplied at beginning of culture, but increased by supplying after 30 days of culture. The results of dynamics indicated that fresh and dry weight of adventitious roots increased with culture period ranged from 0 to 30 days of culture, but did not change from 30 to 40 days;ginsenoside contents did not obviously differed throughout. Sucrose concentration in the culture medium decreased with culture period, and did not exhaust after 40 days culture when the medium supplemented with 50 g/L sucrose at the beginning. Furthermore, pH value was decreased at the initial stage of culture, then gradually increased with culture period, and finally was close to the beginning value.
