摘 要:目的研究虫草肾茶胶囊对培养在高糖条件下系膜细胞(HMC)分泌转化生长因子β1(TGF-β1)及细胞外基质(ECM)的影响,探讨其在糖尿病肾病(DN)防治中的意义。方法应用血清药理学方法,制备虫草肾茶胶囊及福辛普利药物血清,利用酶联免疫吸附试验(ELISA)测定经体外培养的HMC在各种处理因素的作用下上清液中TGF-β1、纤维连接蛋白和层粘连蛋白的表达情况,并以福辛普利为对照。结果高糖能明显促进HMC分泌TGF-β1、纤维连接蛋白和层粘连蛋白(P<0.05或P<0.01)。而虫草肾茶胶囊能抑制HMC分泌TGF-β1、纤维连接蛋白和层粘连蛋白,并呈一定量效关系(P<0.05或P<0.01),且某些方面明显优于福辛普利。结论高糖可促进HMC TGF-β1及ECM分泌,虫草肾茶胶囊能明显抑制高糖条件下HMC分泌TGF-β1及ECM,有助于预防糖尿病肾小球硬化的发生和发展。Objective To investigate the effect of Chongcaoshencha capsule on the expression of TGF-β1and ECM of human mesangial cell(HMC) in vitro under high glucose.To explore the nephroprotective mechanism of Chongcaoshencha capsule for DN.Methods We first get the rats serum with different concentration of Chongcaoshencha capsule and fosinopril.Enzyme linked immunoadsordent assay(ELISA) was adopted to determine the expression of TGF-β1,fibronectin and laminin in cultured human mesangial cells,and the Fosinopril Sodium group was control.Results In vitro,the expressions of TGF-β1,fibronectin and laminin were obviously promoted under the stimulation of high glucose(P〈0.05 or P〈0.01);but Chongcaoshencha capsule can inhibit HMCs secret to TGF-β1,fibronectin and laminin in a dose effect manner(P〈0.05 or P〈0.01),and in some respect the Chongcaoshencha capsule was more effective than the Fosinopril Sodium.Conclusion High glucose can promote secretion of TGF-β1,fibronectin and laminin by HMC,but Chongcaoshencha capsule could inhibit them,which could be helpful for preventing diabetic nephropathy.
