摘 要:2009年河北保定和邯郸地区番茄受到一种毁灭性的新病毒危害,对两地5个病样检测,结果均为番茄黄化曲叶病毒,但未检测到卫星DNAβ分子。对保定和邯郸分离物DNA-A全长进行克隆测序,两分离物DNA-A全长均为2 781 bp,这两个分离物DNA-A全长与其他地区番茄黄化曲叶病毒分离物同源性为98.6%~99.8%,发现其基因间隔区变异稍大,与其他地区番茄黄化曲叶病毒分离物同源性为96.2%~99.7%。邯郸分离物与山东分离物(SD-2)亲缘关系最近,保定分离物与南京分离物(JSNJ1)亲缘关系最近。聚类分析表明,中国几个省市分离物有很高的同源性,但可以分为两个分支,邯郸和保定的分离物分别在两个分支内。A new virus disease occurred in Handan and Baoding district in 2009 which caused heavy loss to tomato production.Five samples showing TYLCV-like symptom were collected and the PCR detection result showed that they were tomato yellow leaf curl virus.The DNAβ were not detected from all the samples.Comparison of DNA sequence of Handan and Baoding isolate showed the complete DNA-A sequence of Handan and Baoding were 2 781 bp and shared high sequence identity with other isolates from China between 98.6% and 99.8%.The intergenic region had the highest diversity compared to other regions and shared sequence identity with other isolates from China between 96.2% and 99.7%.Handan isolate was most closely related to Shandong isolate(SD-2),while Baoding isolate was most closely related to the Nanjing isolate(JSNJ-1).Phylogenic analysis based on the DNA-A sequence showed TYLCV isolates from China were classed into two branches and Handan and Baoding isolates belonged to different branches.
