嗜热菌P4的筛选、鉴定及其高温蛋白酶性质研究

摘　　要：用涂布平板法从温泉水中分离到1株产高温蛋白酶的嗜热菌,命名为P4.通过对菌株P4的生理生化试验和16S rRNA基因鉴定,初步确定其为一株嗜热脂肪地芽孢杆菌(Geobacillus stearothermophilus).菌株P4所产蛋白酶的最适反应温度在65-75℃之间,最适催化pH值为8.0,Zn2+、Mg2+对菌株P4的酶活力具有一定的促进作用.实验结果显示,菌株P4蛋白酶属于嗜热碱性蛋白酶,其酶活力在最适催化温度时达到每毫升发酵液10.0 U.A thermophilic bacterium named P4 was isolated from the hot spring samples. The strain P4 is a Geobacillus stearothermophilus, which was determined by the physiological and biochemical tests and 16S rRNA gene sequence. The optimum reaction temperature was between 65℃ to 75 ℃ and the optimum reaction pH was 8.0. Besides, Zn2＋ and Mg2＋ accelerate the activity. The result shows that the protease of P4 belongs to thermophilic alkaline protease. The enzyme activity in optical catalytic temperature could reach 10.0U per milliliter fermentation fluid.
