摘 要:大环内酯类抗生素因疗效好、安全性高,是临床应用广泛的一类抗菌药物,用于感染性疾病治疗已有50多年的历史。随着耐药菌的日趋常见,医药研究人员加快了对各类新抗微生物药物的研发。其中,通过对大环内酯类抗生素的结构修饰,获得了第三代大环内酯类抗生素——酮内酯类抗生素,已上市的泰利霉素和正在研发的赛红霉素等均显示出良好的应用前景。本文简要回顾大环内酯类抗生素的发展史,着重介绍酮内酯类抗生素的构效关系及其代表性药物的研究近况。As one group of the most widely used antimicrobial agents,macrolides have been applied to the treatment of infectious diseases for more than 50 years owing to their good efficacy and high safety.The research and development for all kinds of new antimicrobial drugs have been accelerated with the rapid increase of drug-resistant pathogenic bacteria.Ketolides the third generation macrolides were obtained by structural modification,in which telithromycin in the market and cethromycin in research and developmenthave shown good prospects in clinic application.History of macrolide development is briefly reviewed.The structure-function relationship of ketolides,the recent progress of the representatives and the other third generation macrolides are mainly introduced in this article.
