高效液相色谱法测定银翘解毒颗粒中绿原酸的含量

摘　　要：目的建立银翘解毒颗粒中绿原酸的HPLC测定方法。方法采用反相高效液相色谱法,色谱柱：DIONEX-ACCLAIM.120 C18（250mm×4.6mm5,μm）;以磷酸盐缓冲溶液（pH=2.5）-甲醇为流动相流速为1.0 mL/min;柱温为35℃;检测波长为350nm。结果绿原酸的线性范围为0.024μg-0.384μg/mLr,=0.9999,平均加样回收率为93.56%,RSD=1.15%（n=6）。结论该方法准确,重复性好,专属性高,适用于银翘解毒颗粒的质量控制。Objective To establish the method for determining Chlorogenic acid in Yinqiao Jiedu granules by RP-HPLC.Methods The HPLC conditions with DIONEX-ACCLAIM.120 C18 column（250mm×4.6mm,5μm）was used,the mobile phase was composed of methy1 cyanide and and PBS（pH=2.5） with gradient elution;flow rate was 1.0mL/min;column temperature was 35℃;the detective wavelength was 350nm.Results The linear ranges of Chlorogenic acid was 0.024μg-0.384μg/mL;r=0.9999.The average recoveries（n=6） of Chlorogenic acid was 93.56%;RSD was 1.15%.Conclusion The method is accurate,reproducible and highly selective,thus suitable for quality control of Yinqiao Jiedu granules.
