摘  要:目的建立喷昔洛韦乳膏体外透皮接收液HPLC测定的方法,研究喷昔洛韦乳膏的体外透皮能力。方法采用Hypersil BDS C18色谱柱(200 mm×4.6 mm,5μm),,流动相:0.02mol/L磷酸二氢钾溶液-甲醇(98:2);流速:1.0mL/min;检测波长:254nm。体外透皮实验采用改良的Franz扩散池,以实验用离体乳猪皮为透皮屏障,自制乳膏与市售乳膏的透皮结果对比,分别在不同时间点取样测定透皮接收液中喷昔洛韦浓度,计算累积透皮量。结果喷昔洛韦的标准曲线方程为A=24435.63C-181.02(r=0.9999),线性范围0.1-1.0μg/mL,平均回收率为98.19%。累积透皮吸收量与时间呈良好的线性关系,自制乳膏与市售乳膏的透皮结果差异无显著性。结论该检测方法操作简便,快速准确,是考察喷昔洛韦乳膏体外透皮性能的较理想的方法。Objective To establish an HPLC method for quantitative determination of penciclovir in vitro percutaneous solution,and to investigate the ability of percutaneous penetration of penciclovir cream.Methods Hypersil BDS C18 column(200mm×4.6mm,5μm) was used as the fixed phase and a mixture of 0.02 mol/L potassium dihydrogen phosphate solution and methanol(98:2) as a mobile phase.The flow rate was 1.0 mL/min.The detection wavelength was at 254 nm.Improved Franz type diffusion cells were used in vitro permeation studies and excised minipigs skins in vitro were used as transdermal barrier.The concentration of penciclovir in the receptor solution was determined by HPLC to investigate its cumulative permeation quantities at different time respectively and compared with that of the commercial cream.Results The regression equation of penciclovir was linear in the range of 0.1-1.0μg/mL(A= 24435.63C-181.02,r=0.9999).The average recovery rate was 98.19%.A good linear relationship existed between the cumulative penetration quantities and the time.There was no significant difference in the permeation quantities between the self-made and commercial cream.Conclusion The method was simple,rapid,and accurate.It is a better method to evaluate the characteristic of permeation for penciclovir.
