摘 要:类黄酮化合物是植物次级代谢产物,参与诸如植物色素沉积、紫外线防御、抵抗病虫害以及花粉发展等多个植物生长和发育过程.作为类黄酮生物合成途径中的关键酶,查尔酮异构酶(CHI,EC 5.5.1.6)在被子植物中广泛存在.为了进一步理解编码查尔酮异构酶的基因家族在被子植物中的整体进化规律,我们利用NCBI和TAIR数据库对被子植物查尔酮异构酶基因家族的相关基因进行了检索分析,共获得522条氨基酸编码序列,去除冗余排列和氨基酸小片段,113条用于系统发育分析.NJ树将CHI氨基酸划分为5个分支,其中分支I包括多数被子植物,分支II只包括豆科植物,分支III、IV和V尽管包含的植物种类较少,但其序列与一些真菌和细菌的CHI蛋白质的保守序列区存在较大相似性.系统分析的结果表明chi基因起源古老,目前的分布现状是基因不断复制和丢失的动态结果.Flavonoids are plant secondary metabolites,they involved in various biological processes during plant growth and development,including pigment accumulation,resistance against pathogenic microorganisms and insect pests,and pollen development.Chalcone isomerase(CHI,EC 5.5.1.6) is an important enzyme in the flavonoid biosynthesis pathway,and exists extensively in angiosperm.In order to further understand the general evolutionary regulation of CHI gene family in angiosperm,522 amino acid sequences were retrieved from NCBI and TAIR databases,and 113 of them were used for phylogenetic analysis after removing redundancies and fragments.The NJ tree grouped the 113 sequences to 5 clusters.Cluster I included many of plants in angiosperm,Cluster II had only some plants in family Leguminaceae,and though Cluster III,IV and V possessed less plants,the sequences shared higher homology with conservative regions of genes from some fungi and bacilli.The present phylogenetic analysis implied that chi genes had old origin and experienced a dynamic process with gene duplication and deletion.
