己烯雌酚阳离子牛血清白蛋白免疫原的合成及鉴定

摘　　要：目的合成阳离子牛血清白蛋白(cBSA),制备多克隆抗体,为动物性食品安全快速检测提供技术支持。方法合成免疫原(DES-HS-BSA和DES-HS-cBSA),采用SDS-PAGE电泳和质谱法对其鉴定,Bradford试剂盒对其定量,经动物免疫制备多克隆抗体,ELISA测定抗体效价和抗体灵敏度。结果成功合成己烯雌酚免疫原,4次免疫后,DES-HS-cBSA组小鼠血清中针对DES-HS的抗体效价(1∶12 000)明显高于DES-HS-BSA组小鼠(1∶4000)。结论 cBSA能增加与小分子结合时的偶联比,作为免疫原的载体蛋白能明显提高抗体效价。Objectiv To synthesize cationized bovine serum albumin（cBSA） and to prepare polyclonal antibody,so as to provide technical support for rapid detection of animal food safety.Methods Diethylstilbestrol hemisuccinate（DES-HS） was reconstructed,and immunogen（DES-HS-BSA and DES-HS-cBSA） was synthesized by EDC method.The results were identified by mass spectrography and SDS-PAGE,and quantitated by Bradford kit.The polyclonal antibody was obtained by animal immunization.ELISA was used to characterize the antibody.Results The immunogens of DES were synthesized successfully.The titre of DES-HS-cBSA group（1∶12 000） was obviously higher than that of DES-HS-BSA group（1∶4000） after four times of immunization.Conclusion The conjugation ratio can be increased when cBSA binding to hapten,instead of BSA.The titre of antibody can be improved as cBSA is the carrier protein of immunogen.
