摘 要:目的研究黄芪多糖(Astragalus Polysaccharides,APS)对糖尿病足溃疡(diabetic foot ulcers,DFUs)成纤维细胞(fibroblasts,Fb)增殖和胶原合成的影响,并探讨其与溃疡愈合的关系。方法取非糖尿病创面及糖尿病足溃疡皮肤进行Fb培养后,分为对照组、DFUs组及APS(10、40、160、640μg/mL)组,观察Fb增殖及胶原合成的变化。结果与对照组相比,糖尿病足溃疡处Fb增殖、胶原合成能力减退;APS在10~160μg/mL浓度范围内可促进体外培养的糖尿病足溃疡Fb增殖,增加胶原合成,且呈量效、时效依赖关系;过高浓度(≥640μg/mL)的APS对Fb增殖、胶原合成产生抑制作用。结论在一定浓度范围内APS促进糖尿病足溃疡Fb增殖和胶原合成,过高浓度APS则产生抑制效应。Objective To investigate the influence of Astragalus Polysaccharides(APS) on the proliferation and collagen synthesis of fibroblasts from diabetic foot ulcers(DFUs).Methods Fibroblasts were isolated and cultured in vitro,and then exposed to different consistencies of APS(10,40,160,640 μ g/mL) respectively.The proliferation of fibroblasts was determined by MTT and the collagen synthesis was measured with the method of hydroxyproline.Results Compared with control group,the proliferation and collagen synthesis of fibroblasts derived from DFUs decreased.APS(10-160 μ g/mL)groups could promote it in a time-dose manner.APS 640 μg/mL groups could inhibit it. Conclusion APS could promote the proliferation and collagen synthesis of fibroblasts from DFUs within a certain range of consistencies,over-high consistency of APS might inhibit it.
