缬沙坦对2型糖尿病大鼠心肌细胞凋亡的影响

摘　　要：目的探讨缬沙坦对糖尿病大鼠心肌细胞凋亡的作用。方法随机选择29只健康雄性SD大鼠中的21只,以高糖高脂饲料喂养4周后腹腔注射链脲佐菌素30mg/kg,3d后测血糖≥16.7mmol/L者(n=19)入选糖尿病大鼠模型,并随机分为糖尿病组(DM组,n=9)和缬沙坦治疗组(VAL组,n=10),另外8只健康雄性SD大鼠为对照组(CN组,n=8)。VAL组给予缬沙坦(20mg/kg.d)治疗6周,检测各组大鼠血生化和胰岛素水平,各组大鼠于第12周末处死,取部分左心室前壁组织以TUNEL法检测大鼠心肌细胞凋亡,免疫组化法检测Caspase-3、NF-κB的表达。结果与CN组相比,DM组和VAL组大鼠心肌细胞凋亡及Caspase-3和NF-κB的阳性表达显著增多,差异具有统计学意义(P<0.05);与DM组相比,VAL组大鼠心肌细胞凋亡及Caspase-3和NF-κB的阳性表达明显减少,差异具有统计学意义(P<0.05)。结论缬沙坦能够抑制糖尿病大鼠心肌细胞的凋亡,具有心肌保护作用。Objective To investigate the effect of valsartan on myocardial apoptosis in diabetic rats.MethodsOf 29 healthy male SD rats,21 were selected and fed high-fat and high-sucrose diet for 4 weeks,and then were injected intraperitoneally streptozotocin（STZ,30 mg/kg）.After 3 days,the rats with plasma glucose≥16.7 mmol/L were chosen for establishing the rat model of diabetes and were randomly divided into the diabetes group（DM group,n=9） and valsartan group（VAL group,n=10）.Other 8 healthy male SD rats were included into the control group（CN group,n=8）.VAL group was treated with valsartan（20 mg/kg.d） for 6 weeks.The biochemical test and insulin level were detected in rats of all groups.At the end of the 12th week the rats of all groups were killed and the part of the left ventricular anterior wall tissue were taken for detecting myocardial apoptosis by TUNEL assay and expressions of caspase-3 and nuclear factor-kappa B（NF-κB） by immunohistochemistry.Results The myocardial apoptosis and positive expressions of caspase-3 and NF-κB increased significantly in DM group and VAL group compared with CN group（P0.05）.The myocardial apoptosis and positive expressions of caspase-3 and NF-κB decreased significantly in VAL group compared with DM group（P0.05）.Conclusion Valsartan can inhibit the myocardial apoptosis and protect the myocardium in diabetic rats.
