摘 要:目的探讨基因的联合干扰对人胃癌SGC-7901细胞增殖的影响。方法将转染细胞分为共四组:psi-control对照组(A组)、psi-端粒酶催化亚基(hTERT)1组(B组)、psi-hTERT1和psi-hTERT2共转染组(C组)、psi-hTERT1和psi-端粒酶调节相关蛋白(TRAP)1共转染组(D组)。采用RT-PCR及Western blot分别检测hTERT mRNA和蛋白表达,端粒重复序列扩增-PCR法检测端粒酶活性,MTT法检测SGC-7901细胞增殖。结果与A组相比,B组、C组hTERT mRNA和蛋白表达、端粒酶活性降低(P<0.05),细胞增殖减慢,细胞凋亡增加;而D组各指标与A组比较差异无统计学意义(P>0.05)。结论联合干扰的效果可能与剂量和目的基因有关。Objective To investigate the effect of combined interference of genes on prolifera-tion of human gastric cancer SGC-7901 cell.Methods The transfected cells were divided into four groups of A(transfected with psi-control as the control),B[transfected with psi-human telomerase catalytic subtmit(hTERT)1],C(co-transfected with psi-hTERT1 and psi-hTERT2) and D [co-transfected with psi-hTERT1 and telomerase regulation-associated protein(TRAP)1].The expressions of hTERT mRNA and protein were detected by RT-PCR and Western blot,respectively.The activity of telomerase was measured by telomeric repeat amplification protocol-PCR method and the cell proliferation was examined by MTT assay.Results Compared with group A,the expressions of hTERT mRNA and protein and the activity of telomerase were decreased(P0.05),the cell proliferation was declined and the cell apoptosis was increased in groups of B and C.However,there was no significant difference of each index between group D and group A(P0.05).Conclusion The effect of combined interference may be related with the dose and the target gene.
