摘  要:目的:建立复方土茯苓颗粒的提取、纯化工艺。方法:采用正交试验优化复方土茯苓颗粒提取、醇沉工艺;采用HPLC法测定提取物中牛膝含有的蜕皮甾酮。结果:确定提取工艺为第1次用8倍量水煎煮1h,第2次用7倍量水煎煮1h;醇沉工艺为将药液浓缩至0.5g/mL(相当于药材),边搅拌边加适量乙醇至浓度为60%,静置12h,分离上清液。结论:验证试验表明,该提取、醇沉工艺客观可行、稳定合理,可为工业化生产提供理论依据。Objective:To optimize the extracting and alcohol precipitating craft of compound Tufuling granules by Orthogonal experiment.Methods:Orthogonal experiment was applied to optimize the influence factors of extracting and alcohol precipitating;ecdysterone in Radix Achyranthis Bidentatae of compound Tufuling granules concrete was determined by HPLC.Results:The best extraction craft is that the herbs should be extracted by 8 times(V/V) water for 1h,then be extracted by 7 times water for 1h;The best alcohol precipitating craft is the concrete should be concentrated to 0.5g/mL(counted by herbs),added alcohol to 60%(V/V) with vigorously stirring,then placed for 12h and centrifugalize the supernatant fluid.Conclusion:Proving trial showed that this extracting and alcohol precipitating craft is scientific and reasonable,and can offer theory basic for industrialized manufacture of compound Tufuling granules.
