摘 要:目的:研究白芷提取物对酪氨酸酶的抑制作用。方法:将白芷的95%乙醇提取物依次用石油醚、乙酸乙酯、正丁醇萃取,并用曲酸作阳性对照、以酪氨酸作底物来测定石油醚、乙酸乙酯、正丁醇萃取部分及萃取后水部分对酪氨酸酶的抑制作用。将白芷乙酸乙酯部分进一步用正相硅胶柱和Sephadex LH-20凝胶柱分离纯化得到四个单体化合物,分别为丁二酸、反式咖啡酸、水合氧化前胡素和白当归脑,并对4个单体化合物进行酪氨酸酶抑制试验。结果:经酪氨酸酶活性测定,白芷乙酸乙酯萃取部分对酪氨酸酶的活性抑制作用较强,其半数抑制浓度IC50为1.25 mg/mL,白芷石油醚部分和萃取后水部分均无抑制作用,而白芷正丁醇部分相反有激活酪氨酸酶活性作用。除反式咖啡酸有较弱的酪氨酸酶活性抑制作用外,其它3个单体化合物均无抑制作用。结论:白芷乙酸乙酯部分有较明显的酪氨酸酶抑制作用。本研究没有获得具有明显抑制酪氨酸酶活性的单体化合物,有待于进一步分离纯化研究。Objective:To study on tyrosinase inhibitory activites of extracts from Angelica dahurica.Methods:The 95% alcohol extract from Angelica dahurica was suspended with water,and then extracted with petroleum ether,ethyl acetate and n-butanol in proper order,respectively.Using Kojic acid as the positive control and L-tyrosine as substrate,the petroleum ether,ethyl acetate,n-butanol and residue water extracts of Angelica dahurica were assayed for tyrosinase inhibitory effects.The active ethyl acetate extract was separated by silica gel and then Sephadex LH-20 column chromatography to give four compounds,which were elucidated as succinic acid,trans-caffeic acid,oxypeucedanin hydrate and byakangelicol respectively.Moreover the four compounds were assayed for tyrosinase inhibitory effects.Results:The ethyl acetate extract of Angelica dahurica showed inhibitory effects on tyrosinase activity and the IC50 value was 1.25 mg/mL.The petroleum ether and residue water extracts of Angelica dahurica have no inhibitory effects,while n-butanol extract showed increasing effect on tyrosinase activity.Among the four compounds,only trans-caffeic acid has weaker inhibitory effects,and the other three compounds have no inhibitory effects.Conclusion:The ethyl acetate extract of Angelica dahurica showed inhibitory effects on tyrosinase activity,but no compound with obvious tyrosinase inhibitory effects was obtained from the ethyl acetate extract of Angelica dahurica.We need to further study on separation and purification of Angelica dahurica.
