摘 要:目的:观察康脑液对脑缺血再灌注大鼠血管新生影响。方法:60只雄性SD大鼠随机分为正常组、模型组、康脑液大剂量组、康脑液中剂量组、康脑液低剂量组,每组12只。线栓法制备大鼠右侧大脑中动脉闭塞(Middle cerebral artery occlusion,MCAO)模型。大鼠缺血2h,再灌注7天,每组取6只大鼠于再取脑TTC染色观察梗死体积,另6只取脑观察各组大鼠脑血管分布情况,制备脑片,免疫组化方法标记CD105观察新生血管密度(MVD)。结果:与模型组相比,康脑液治疗组脑梗死体积明显小于模型组(P<0.05或P<0.01),脑表面血管分布数目明显多于模型组、CD105显著增加(P<0.05或P<0.01)。结论:康脑液能促进脑缺血再灌注大鼠缺血周围区域血管新生,明显减小脑梗死体积。Objective:To observe the efects of kangnaoye on cerebral infarction volume and tissue angiogenesis in rats with cerebral ischemia-reperfusion injury.Methods:60 healthy male SD rats were randomly divided into 5 groups:normal group,model group,high-dose kangnaoye group [28.6g/(kg·d)],middle-dose kangnaoye group [14.3g/(kg·d)] and low-dose kangnaoye group [7.15g/(kg·d)].The model of focal cerebral ischemia and reperfusion was made by thread embolism method,ischemia for 2h,and reperfusion for 7 days.Infarct volume was detected by triphenyltetrazolium chloride(TTC) staining.We observe the blood vessel on the surface of the other six rats and immunohistochemical method was used to investigate the expression of CD105.Results:Compared with model group ones,the cerebral infarct volume of rats in Kangnaoye treatment groups decreased obviously,the number ofblood vessel and the MVD is much higher in Kangnaoye treatment groups(P0.05 or P0.01).Conclusion:kangnaoye can promote tissue angiogenesis in rats with cerebral ischemia-reperfusion injury and protect the neurocyties.
