青蒿琥酯对人白血病K562细胞NF-κB活化的影响

摘　　要：目的:观察青蒿琥酯对白血病细胞K562核转录因子κB信号通路的影响。方法:采用Western blot法,检测青蒿琥酯对K562细胞IKKα,IKKβ,IKKγ)表达的影响,采用Western blot法,检测青蒿琥酯对K562细胞caspase 3和PARP cleavage的作用;采用EMSA法检测青蒿琥酯对白血病细胞NF-κB活性的影响。结果:青蒿琥酯能抑制核转录因子κB的表达,青蒿琥酯能够下调IKKα的表达。结论:青蒿琥酯通过下调IKKα的表达而抑制转录因子κB活化。Objective：To investigate the effects of Artesunate on inhibition the NF-κB activity in K562 cells.Methods：A Western blot was performed to investigate the effect of Artesunate on the expression of IKKα,IKKβ,IKKγ.Western blot was performed to investigate the effect of Artesunate on the expression of caspase 3 and PARP cleavage,EMSA was performed to evaluate the effect of Artesunate on NF-kB activation.Results：Artesunate at pharmacological concentration can inhibit the NF-kB activity.Artesunate can downregulate the NF-kB activator IKK.Conclusion：Artesunate inhibits NF-kB activation mainly through downregulation of the NF-kB activator IKKα.
