摘 要:目的:探讨左金丸的主要成分吴茱萸碱和盐酸小檗碱对结肠癌HT29细胞凋亡及端粒酶催化亚基(hTERT)表达的影响,为临床应用左金丸治疗大肠癌提供实验依据。方法:体外培养人结肠癌HT29细胞,加入不同浓度的吴茱萸碱及盐酸小檗碱进行培养,采用活细胞计数试剂盒(CCK-8)检测细胞增殖,TRAP法检测端粒酶活性,RT-PCR法检测hTERT的表达。结果:吴茱萸碱和盐酸小檗碱作用HT29细胞后,增殖明显受到抑制,呈剂量和时间依赖性;能够降低HT29端粒酶活性,但二者无协同作用;并且端粒酶hTERT表达降低,具有剂量依赖关系。结论:吴茱萸碱和盐酸小檗碱对结肠癌HT29细胞具有显著生长抑制作用,该抑制作用可能通过下调端粒酶hTERT基因表达,降低端粒酶的活性,为临床治疗大肠癌提供实验依据。Objective:To study the effects of evodiamine and berberine hydrochloride on telomerase activity of the cultured human colorectal cancer cell line HT29 and expression of hTERT-mRNA.Methods:HT-29 cells were treated with evodiamine and berberine hydrochloride at different concentrations.The proliferative status of HT-29 cells were measured by WST-8 assay.Telomerase activity was detected by TRAP and hTERTmRNA expression was examined by RT-PCR.Results:Evodiamine and berberine hydrochloride displayed a significant growth inhibitory effect on HT29 cell line in the dose and time-dependent manner.they could inhibit the telomerase activity of HT29 cells,but have no synergistic reaction.Moreover the expression of hTERT-mRNA was decreased in concentration-dependent manner.Conclusion:Evodiamine and berberine hydrochloride can inhibit the proliferation of colorectal cancer cell line HT-29,whose mechanisms may be related to down-regulation of hTERT gene expression and then decreased telomerase activity.
