摘  要:[目的]筛选苹果砧木耐盐变异体,并对其生理生化和分子生物学特征进行分析。[方法]用60Co-γ对苹果砧木78-48进行辐射处理,以处理明显的叶片为外植体诱导出再生植株,以1.0%和1.2%NaCl作为选择压,对再生植株进行多代交替盐筛选,直到筛选出耐1.2%NaCl的变异体L和K,以耐盐变异体L2和K2为试材,对其进行耐盐生理生化指标测定及RAPD分析。[结果]变异体不仅在生理生化指标上有明显的差异,而且在DNA水平上发生了突变。[结论]变异体L2和K2是78-48的耐盐新株系。[ Objective ] The research aimed to screen the salt- tolerant mutant of apple rootstock, and analyze their physiological- biochemis-trical and molecular biological characteristics. [ Method ] The original line 78-48 (CK) was irradiated by ^60Co-y, and the effect obvious leaves were chose to induce the regenerative plants, thenthe plants were alternating salt screened taking 1.0% and 1.2% NaC1 as selection press, until to screen the mutants to 1.2% NaC1- tolerance. Finally, the physiological- biochemistrical indices of the salt- tolerant mutants, L2 and K2, were determined, and their molecular biological characteristics were studied by RAPD analysis. [ Result] The result showed that com- pared with the original line, the mutants varied not only on the salt-tolerant indexes level, but also on the DNA level, which provided a strong evidence for the truth of the mutants. [ Conclusion] These mutants (L2 and K2) were sah-tolerant new lines of apple rootstock.
