摘 要:[目的]建立牛胎肺间充质干细胞(Mesenchymal stem cells,MSCs)体外培养体系并对其进行生物学特性研究。[方法]取3~5月龄牛胎儿肺脏,得到MSCs。通过控制胰酶消化时间,去除难消化的上皮样贴壁细胞,获得纯化的牛肺MSCs,研究其形态、增殖情况及体外诱导分化潜能。[结果]牛胎肺MSCs可在体外扩增培养,传至24代以上,表达CD29、CD44和CD166,不表达CD34、CD45和BOLA-DR,并具有向成骨细胞诱导分化的潜能。[结论]从牛胎肺中可以分离出MSCs,可以在体外培养,并可诱导分化为成骨细胞。[ Objective] The aim was to study on the biological characteristics of bovine fetal lung mesenchymal stem cells (MSCs) by isola- ting culture in vitro. [ Method ] The 3 -5 months lung of bovine fetal was stripped. MSCs were isolated and cultured. By controlling the time of trypsin digestion, MSCs were purified and epithelial - like ceils that were digested difficultly and still adherent was deleted. Then the mor- phology and the growing proliferation condition and the differentiation potential in vitro of the bovine lung MSCs were observed. [ Result] Bo- vine fetal lung MSCs can be cultured in vitro and have been passed more than 24 generation. The MSCs expressed CD29, CD44 and CD166, but didnt express CD34, CD45 and BOLA-DR. Results displayed that the MSCs had the differentiation potential to osteoblasts. [ Conclusion] MSCs from bovine fetal lung can proliferate rapidly in vitro and be induced to asteoblast.
