农药二甲戊乐灵对FRTL-5细胞甲状腺过氧化物酶基因的影响

摘　　要：[目的]观察二甲戊乐灵(Pendimethalin)对FRTL-5细胞甲状腺过氧化物酶基因(thyroperixidase,Tpo)的影响及其机制。[方法]0.1μg/ml,0.5μg/ml和1μg/ml二甲戊乐灵处理FRTL-5细胞后,用MTT法和3H掺入法测细胞毒性;免疫细胞化学法观察二甲戊乐灵对FRTL-5细胞甲状腺球蛋白(thyroglobulin,Tg)和甲状腺促转录因子1(thyroid transcription factor1,TTF-1)的影响;RT-PCR观测二甲戊乐灵对tpo基因的影响。[结果]二甲戊乐灵对FRTL-5细胞无显著细胞毒性;1μg/ml二甲戊乐灵显著增强胞浆内的Tg和tpo基因的转录,0.5μg/ml和1μg/ml二甲戊乐灵显著降低TTF-1表达。[结论]二甲戊乐灵可直接促进tpo基因的转录;同时,二甲戊乐灵可显著增加胞浆内的Tg。[Objective] To investigate the effects of pendimethalin on the thyroperixidase gene in FRTL-5cells.[Methods]FRTL-5cells treated with 0.1μg/ml,0.5μg/ml and 1μg/ml pendimethalin,and cytotoxicity was tested by MTT and 3H-TdR.The effects of pendimethalin on Tg and TTF-1 in FRTL-5 cells were analyzed by ICC.RT-PCR was used to determine the influence of pendimethalin on tpo gene.[Results]Pendimethalin had no significant cytotoxicity to the FRTL-5 cells.The concentration of Tg in the cells and tpo transcription was increased by 1μg/ml pendimethalin,but the TTF-1 was decreased by 0.5μg/ml and 1μg/ml pendimethalin.[Conclusion]Pendimethalin may directly promote the tpo gene transcription,and may also increase the Tg in FRTL-5 cells.
