摘 要:目的探讨补肾养肝熄风汤对帕金森病(PD)小鼠纹状体神经递质和多巴胺(DA)受体的调节机制。方法将C57/BL雄性小鼠56只随机分为正常组、模型组、中药组和西药组,每组14只。采用MPTP腹腔注射复制PD小鼠模型,正常组、模型组生理盐水灌胃(0.1ml/10g),中药组以补肾养肝熄风汤灌胃(0.1ml/10g),西药组多巴丝肼灌胃(0.1ml/10g),每日1次,治疗14d。治疗结束后分离取出小鼠纹状体组织,测定纹状体DA、去甲肾上腺素(NE)、5-羟色胺(5-HT)、亮胺酸脑啡肽(LEK)含量以及DAD1、D2受体的含量。结果模型组、中药组和西药组小鼠纹状体DA、NE、5-HT、LEK含量显著低于正常组(P<0.05或P<0.01),中药组和西药组DA、NE、5-HT含量显著高于模型组(P<0.05或P<0.01);模型组、中药组和西药组纹状体DAD1受体免疫组化指数(IHC)显著低于正常组(P<0.01),西药组DAD1受体IHC高于模型组和中药组(P<0.05);中药组、西药组DAD2受体IHC显著高于模型组(P<0.01)。结论补肾养肝熄风汤可提高纹状体下降的DA、NE、5-HT水平,可提高纹状体DAD2受体的含量。Objective To observe the adjustment mechanism of Bushen Yanggan Xifeng Decoction (Decoction for reinforcing the kidney, nourishing the liver, and stopping wind) on neurotransmitters and dopamine receptor in the striatum of mice with parkinson's disease (PD). Methods Fifty-six adult male C57/BL mice were randomized into normal group, PD model group, TCM group, and western medicine group, 14 in each. The PD models were established by intraperitoneal injection of MPTP. The normal group and model group were given intragastric feeding with normal saline (0. 1ml/10g), the TCM group with Bushen Yanggan Xifeng Decoction (0. 1ml/10g), and the western medicine group with Levodopa and Benserazide Tablet (0. 1ml/10g), once every day, for 14 days. The level of striatal DA, NE, 5-HT, level of striatal LEK and content of dopamine receptors, D1R and D2R in each group were assayed. Results The levels of striatal DA, NE, 5-HT, and LEK in the model group, TCM group, and western medicine group were lower significantly than those in the normal group (P〈0.05 or P〈0.01), and the levels of DA, NE, and 5-HT in the TCM group and western medicine group were higher significantly than those in the model group (P〈0.05 or P〈0.01). Immunohistoryehemistry (IHC) index of striatal D1R in the model group, TCM group, and western medicine group was lower significantly than that in the normal group (P〈0.01), and the IHC index of striatal D1R in the western medicine group was higher significantly than that in the model group and TCM group (P〈0.05). IHC index of striatal D2R in the TCM group and western medicine group was higher significantly than that in the model group (P〈0.01). Conclusion Bushen Yanggan Xifeng Decoction can increase the lowered striatal DA, NE, and 5-HT of mice models and increase the content of striatal D2R as well.
