摘 要:利用巢式PCR和SSR技术研究了0,15,25,50,75,100,125,150 mg.L-1的硫酸二甲酯(DMS)对白花泡桐幼苗形态、植原体和DNA碱基序列的影响.结果表明,15 mg.L-1DMS处理3 h可使丛枝病幼苗转变为形态上健康幼苗,但其幼苗内仍有丛枝病植原体的存在;25~125 mg.L-1DMS处理可使丛枝病幼苗呈现健康状态,并且幼苗内无植原体存在,125 mg.L-1DMS处理3 h的幼苗生长受到抑制.白花泡桐丛枝病幼苗、健康幼苗、DMS处理后形态呈现健康幼苗及其检测不到植原体存在幼苗的DNA的碱基序列没有发生变化.说明适宜质量浓度DMS可抑制丛枝病植原体,使丛枝病幼苗转变为健康幼苗;泡桐丛枝病发生前后及DMS处理病苗DNA的SSR扩增位点相同.Effects of 0,15,25,50,75,100,125,150 mg·L-1 DMS on the morphological and phytoplasma changes of Paulownia fortunei seedlings with witches' broom and DNA base sequences were studied with nest-PCR and SSR.The results indicated that there existed phytoplasma in the infected seedlings treated with 15 mg·L-1 DMS for 3 h which might turn into healthy ones morphologically,but phytoplasma disappeared in the infected seedlings treated with 25~125 mg·L-1 DMS,and the seedling growth were inhibited with more than 125 mg·L-1 DMS.And the DNA base sequences of the healthy,infected and the infected-treated with DMS seedlings were the same with SSR analysis.The seedlings with witches' broom treated with appropriate concentrations of DMS might become healthy ones.And there existed the same DNA loci at SSR level among the healthy,infected and infected-treated with DMS seedlings.
