摘  要:目的观察E-钙粘蛋白、基质金属蛋白酶-9(MMP-9)在肾间质纤维化(R IF)大鼠肾组织中的表达,探讨E-钙粘蛋白、MMP-9在R IF中的作用。方法选用SD大鼠72只。随机分为正常对照组、假手术组和模型组,制备单侧输尿管梗阻UUO模型。采用病理及免疫组织化学等方法动态观察各组大鼠第1天、第3天、第7天、第14天肾脏组织学变化和E-钙粘蛋白、MMP在肾小管间质中表达情况。结果 UUO模型组大鼠肾小管间质损害明显,肾小管上皮细胞E-cadherin表达逐渐下调,于第14天达最低值。肾小管上皮细胞MMP-9表达上调,第3、7、14天与同期正常对照组、假手术组相比差异显著。肾小管上皮细胞E-钙粘蛋白表达量与肾小管间质损害程度呈负相关,MMP-9表达量与肾小管间质损害程度呈正相关。结论 E-钙粘蛋白表达下调、MMP-9表达上调、E-钙粘蛋白与MMP-9表达失衡可能是肾间质纤维化形成促进因素之一,并可能参与肾间质纤维化形成的全过程。Objective To observe the expression of E-cadherin and matrix metalloproteinase-9(MMP-9) at kidney in rats with renal interstitial fibrosis(RIF),and explore the effects of E-cadherin and MMP-9 in RIF.Methods seventy-two male Sprague-Dawley(SD) rats were randomly divided into normal control group(n =24),sham-operated group(n =24) and model group(n=24).Tubulointerstitial fibrosis model was established via unilateral ureteral obstruction(UUO).Histological changes were observed under microscope and immunohistochemistry staining was performed to investigate the expression of E-cadherin and MMP-9 in RIF by semiquantitative score at 1,3,7 and 14 days after experiment onset.Results Obvious tubulointerstitial injury was observed in UUO model.In model group the expression of E-cadherin in the tubular cells decreased and reached the minimum on day 14;the expression of MMP-9 in the tubular cells increased significantly as compared to the normal group and sham-operated group on days 3,7 and 14.There was negative correlation between renal interstitial injury and E-cadherin expression and positive correlation between renal interstitial injury and MMP-9 expression.Conclusion The lower expression of E-cadherin and higher expression of MMP-9 induced inequality between E-cadherin and MMP-9 may promote the progress of RIF and play an important role in the whole process of RIF.
