锌转运体8在胰岛功能和血糖稳态中的作用

摘　　要：锌转运体8(ZnT8)是新近发现的特异性高表达于胰岛的锌转运体,主要功能是参与锌在细胞内的区室化以及锌的外排,从而降低胞质内锌的浓度。同时,SLC30A8多态性影响2型糖尿病的易患性,与2型糖尿病的发病机制有关。虽然ZnT8全身剔除的小鼠空腹及葡萄糖刺激后胰岛素均减少,但血糖浓度无明显改变,表明了ZnT8的缺乏影响胰岛功能,而对全身血糖代谢的影响比较局限。ZnT8（SLC30A8）is a newly discovered islet-specific zinc transporter that controls zinc efflux into the extracellular matrix and intracellular vesicles to reduce the concentration of zinc in the cytoplasm.The polymorphism of the SLC30A8 gene is associated with susceptibility to type 2 diabetes.ZnT8 deletion decreased fasting and glucose-stimulated insulin secretion,however the blood glucose levels were not significantly changed in mice.In summary,SLC30A8 gene deletion is accompanied by a modest impairment in insulin secretion without major alterations in glucose metabolism.
