摘  要:0 引 言蛋白质的定量是生物化学、临床医学乃至食品检验等最常涉及的内容。所使用的方法多种多样,其中用小分子作为光谱探针测定蛋白质以其简便、灵敏等优点而被广泛应用,如考马斯亮蓝G250[1]、溴酚蓝[2]、氨基黑10B[3]等。但是,这类探针反应的作用机理迄今仍不很清楚...In this paper,it is found firstly that BSA can sensitively react with carboxynitroazo,used as a good colouring reagent of some inorganic ions before,in acid medium and at room temperature.The absorption spectra and its reaction mechanism were studied.BSA binds with carboxynitroazo mainly through the static electric force.The maximum combination number of carboxynitroazo binded on BSA was determined,based on principle of molar ratio method.The results were conform to the change of its spectra.The fundamental conditions of the reaction and the possibility of spectrophotometric determination of proteins using carboxynitroazo were discussed.The determined molar absorptivitys were 2 43×10 5?L·mol -1 ·cm -1 for BSA and 4 48×10 5?L·mol -1 ·cm -1 for γ\|G,respectively.
