摘 要:目的优化黄嘌呤氧化酶法测定大鼠血清SOD实验条件。方法采用黄嘌呤氧化酶法,探讨其加样量、方法重复性、显色稳定性、试剂稳定性及样品稳定性等最佳因素。结果黄嘌呤氧化酶法测定大鼠血清SOD最佳加样量为5μl,其测定变异系数CV<1%,显色前后20 min吸光度比较(P>0.05);新配试剂与配制3个月试剂做测定,吸光度比较(P<0.01);新鲜标本和4°C放置48 h标本进行测定,吸光度比较(P<0.01)。结论黄嘌呤氧化酶法测定大鼠血清SOD最佳加样量为5μl,该法重复性好,显色稳定,测定试剂必须新鲜配制,标本必须新鲜制备。Objective To experimental conditions. optimize the measurement of the serum SOD under the xanthine oxidase Method to use xanthine oxidase to explore the optimal factors such as the amount of the added sample, method reproducibility, color stability, reagent stability and sample stability for the measurement. Result The optimal volume of SOD was 5 μl, the coefficient of variation was CV〈1% and the color absorbance was P〈0.05% 20 minutes before and after the test. Compared with 3-month reagents, the new one was found to have a color absorbance of P〈0.01 and the color absorbance was P〈0. 01% when the new reagent was stored at 4 ℃ for 48 h. Conclusion The optimal sample volume was 5 μl when xanthine oxidase was used to measure serum SOD, which is of good reproducibility with a color stability. The reagent and the specimens are both suggested to be freshly prepared.
